I'm not sure what your question is. You want users inside your company who want to browse the web (port 80) forced to use a proxy server? And you want to do this by using DNAT so that outgoing http traffice (port 80) is rerouted to your proxy server?

Yes. For my question, users doesn't explicit specify proxy address but web traffics redirected to proxy server by ASL.

Cannot you use the integrated HTTP proxy (Squid)  in transparent mode?

Yes, I can't enable transparent proxy feature. When I tried to enable, ASL will not respond with any traffic. With hardware reported, load was too much (>2.0). So, I try to separated proxy function to another servers.

Your ASL system should have no trouble running squid according to your specs.

It does take time for squid to start up, esp the first time you run it.

Nonetheless, if you want to use DNAT, it will work.
I think you'd want:

from INT (any port), to ANY DEST PORT 80, change DEST to PROXY SERVER PORT 8080

and make sure you have a packetfilter rule that allows traffic on 8080 from INT to PROXY
