SMTP Proxy DNS setting

You define your external DNS lookup servers in the DNS proxy (Proxy --> DNS).
If you use your ISP's outbound mail server as a smart host mail forwarder, you can also define it as a host under Definitions --> Networks.

The settings the SMTP proxy uses are the same ones used by the DNS proxy settings.

Turn it on and disable on all interfaces, if you disable the proxy it still uses the settings for internal use.

But what if DNS Proxy is disabled? 

Turn it on ASAP. There is no advantage in leaving it disabled.

On the networks that I look after, we use split horizon DNS. The workstations on the Internal network are given the addresses of our two internal DNS servers (Windows 2003 domain controllers) via DHCP. The two domain controllers forwards DNS requests that they can't resolve to the Astaro DNS proxy. The Astaro DNS proxy has been configured with the three DNS server addresses provided by our ISP. Thus the only DNS requests going out on the WAN, are those coming from the Astaro DNS proxy.

Yes, do turn on the DNS proxy and configure forwarders... THe web proxy, smtp proxy, etc. all use it when it's available, and the web proxy seems to run faster (quicker DNS lookups, I think) when the dns proxy is enabled.
